Рётаро Хашизуме (Ryotaro Hashizume) с коллегами из Высшей школы медицины Университета Миэ (Mie University Graduate School of Medicine) в Японии использовали систему редактирования генов CRISPR-Cas9 для расщепления третьей хромосомы в ранее созданных клеточных линиях с трисомией 21. Модифицированный учеными метод позволяет редактору CRISPR-Cas9 выборочно нацеливаться и устранять дополнительную копию хромосомы 21, вызывающую синдром Дауна, эффективно восстанавливая нормальную клеточную функцию как в стволовых клетках, так и в специализированных клетках человека. Суть нового подхода в том, что редактор идентифицирует и вырезает определенные последовательности ДНК, обнаруженные только на дополнительной хромосоме. Это позволяет удалять только дополнительную копию, оставляя нормальную пару хромосом нетронутой. Исследование опубликовано в журнале PNAS Nexus. В лабораторных экспериментах метод успешно сократил количество хромосом с трех до нормальных двух как в стволовых клетках, так и в клетках кожи, полученных от человека с синдромом Дауна. Скорость исправления достигла почти 17% в стволовых клетках, когда исследователи также временно подавили определенные механизмы восстановления ДНК.
Примечательно, что клетки, у которых была удалена дополнительная хромосома, показали значительные улучшения в нескольких аспектах. Так, в частности, у них были отмечены повышенные темпы роста и снижение выработки вредных активных форм кислорода, представляющее собой известную проблему в клетках с синдромом Дауна. Кроме того, паттерны активности генов также сместились ближе к тем, которые наблюдаются в клетках без синдрома Дауна. Исследователи обнаружили, что исправление числа хромосом восстанавливает нормальные паттерны активности генов и улучшает клеточную функцию. Важно, что новый метод работал не только в стволовых клетках, но и в специализированных клетках, которые не делятся активно. Хотя исследование знаменует собой значительное техническое достижение, путь к любым потенциальным клиническим применениям остается долгим и сложным. Использование нового метода CRISPR-Cas, как и его предыдущих версий, сопряжено со случайными непреднамеренными генетическими модификациями.
